摘  要:目的探讨维生素E对老年小鼠细胞免疫功能及心、脑脂质过氧化反应的影响。方法3月龄幼年小鼠和18月龄老年小鼠各饲以含维生素E(VitE)500×10-6和30×10-6饲料8周后,测定血清VitE水平、脾细胞转化反应和心、脑脂褐质含量。结果饲以含500×10-6VitE饲料的老年小鼠脾细胞对刀豆蛋白A(ConA)和脂多糖(LPS)的反应及血清VitE水平均较饲以含30×10-6VitE饲料者显著增高(P值<0.01);心、脑组织中脂褐质含量则明显降低(分别为P值<0.01及0.05)。结论膳食中补充较高剂量VitE后,能显著提高老年小鼠血清VitE水平,增强脾细胞转化反应,并明显抑制心、脑脂质过氧化。Objective In this study, the effect of vitamin E (Vit E) on cell-mediated immune functions and lipid peroxidation in the brain and heart of aged mice were observed. Methods Semi-purified diet containing 30×10(-6) or 500×10(-6) DL-α-tocopherol (Vit E) was fed for 8 weeks to young (3 months) and old ( 18 months) Swiss mice. The average daily intake of Vit E was 42mg/kg or 2.5mg/kg body weight for the young and 30mg/kg or 1.8mg/kg body weight for the aged. The blastogenic response of splenocytes as well as the serum Vit E and lipofuscin levels in brain and heart were measured. Results It is shown that the old mice fed with 500×10(-6) Vit E diet had a significantly higher serum Vit E level and blastogenic response of splenocytes to Con A and LPS than those fed with 30× 10(-6) Vit E diet (P<0.01). However, lipofuscin level in the brain and heart of the aged mice declined substantially by Vit E supplementation (heart:P<0.01, brain:P< 0.05 ).Conclusions Higher dose of dietary Vit E supplementation significantly enhanced the serum Vit E level and blastogenic response of splenocytes of aged mice,while obviously inhibited lipid peroxidation and thereby reduced lipofuscin accumulation in the tissues.
