摘 要:目的:用三维多孔的β-磷酸三钙做支架材料,犬的骨髓基质细胞(MSCs)为种子细胞,观察它的成骨能力,探讨其修复犬下颌骨3 cm节段性缺损的可行性。方法:3 cm×2 cm×1 cm的三维多孔β-磷酸三钙接种犬的MSCs,然后将支架材料/MSCs复合物移植在犬下颌骨3 cm长的节段性骨缺损处,饲养动物6个月,组织形态学观察成骨能力,并和单纯用支架材料修复组对比。结果:支架材料/MSCs复合物形成大量的组织工程骨,它修复的犬下颌骨缺损区的形态优于对照组。结论:三维多孔的β-磷酸三钙/MSCs复合物形成的组织工程骨,有良好的外部形态,能修复犬下颌骨3 cm长节段性缺损。Objective: To test the feasibility of three dimensional porous β- tricalcium phosphate( β -TCP)scaffold with marrow mesenchymal stem cells (BMSCs) in reconstructing three -centimeter long segmental defect in canine mandible. Methods: Firstly we seeded BMSCs in 3 cm × 2 cm × 1 cm three - dimensional porous β - TCP scaffold, then placed the β - TCP/BMSCs composites in three - centimeter long segmental defects in canine mandibles. The three - dimensional porous β - TCPs alone was used as control. After 6 months, the osteogenic ability of β - TCP/BMSCs composites was assesed by histomorphological method, and compared with that of control group. Results: β - TCP/BMSCs composites formed more bone, and the shapes of the reconstructed canine mandibular defects by β - TCP/BMSCs were better than those of control group. Conclusion: The tissue enginerring bone formed by β -TCP/BMSCs composites has better contour, and it can be used to repair the three - centimeter long segmental defect in canine mandible.
