摘 要:应用PCR-SSCP技术首次分析5个品种鸭生长激素基因座位5’端调控区、外显子与部分内含子和3’端调控区中的多态位点,对具有多态性的位点进行测序,以探讨鸭生长激素基因遗传变异情况,比较不同品种该基因座位遗传多样性差异。结果在5个品种鸭中共检测到5个多态位点,多态位点频率在5个不同品种鸭中存在明显差异。序列比较分析结果表明:鸭生长激素基因座位5’端与3’端调控区和外显子序列具有极高的保守性,在5个鸭品种间不存在任何差异,所测出的序列与已知鸭GH序列相应部分完全一致。检测出的6处突变点(包括4个替换、1个插入和1个缺失)均位于内含子区域。推测鸭生长激素基因表达差异可能受内含子序列影响。DNA polymorphism of growth hormone gene in five duck breeds including 5' flanking, 3' flanking and exon sequence were studied using PCR-SSCP. Five GH alleles were identified in five duck breeds. And their frequencies were significantly different between breeds. There were high conservatism in duck GH gene 5' flanking, 3' flanking and exon sequence and no differences among the five breeds. Meanwhile, there were six mutational sites (4 substitutions, 1 insertion and 1 absence) in intron region.
