摘  要:目的:动态观察大鼠糖尿病肾病发生、发展过程中纤溶酶原激活物抑制物-1(plasminogen activatorinhibitor-1,PAI-1)在肾脏的表达变化,旨在探讨PAI-1在糖尿病肾病(diabetic nephropathy,DN)肾纤维化发病机制中的作用。方法:链脲菌素(streptozocin,STZ)诱导大鼠糖尿病肾病,以免疫组织化学及荧光实时定量PCR(RT-PCR)的方法动态观察糖尿病肾病发生、发展过程中大鼠肾脏PAI-1蛋白及mRNA的表达。结果:PAI-1蛋白表达从糖尿病2周起较正常对照组明显增多(P<0.05),且随着糖尿病进展有逐渐增加的趋势,阳性表达主要位于肾小管上皮细胞的胞浆内。用RT-PCR的方法检测大鼠肾脏PAI-1 mRNA的表达,结果显示16周DN组大鼠肾组织中PAI-1 mRNA的表达显著高于正常对照组大鼠(P<0.05)。结论:PAI-1表达上调可能参与了糖尿病肾病中肾脏纤维化形成的过程。Objective: To study the role of plasminogen activator inhibitor-1 (PAI-1) in development of renal fibrosis in diabetic nephropathy. Methods: Rat diabetic nephropathy was induced by using streptozocin (STZ) tail intravenous injection. The expression of PAI-1 protein and mRNA were examined with immunohistochemical technique and RT-PCR. Results: With the progression of diabetic nephropathy renal fibrosis, the expression of PAI-1 protein and mRNA in kidney of diabetic rats elevated, and in the 16^th week of the experiment, they were significantly higher than those in rats of control group(P 〈0. 05). Conclusions: In STZ-induced diabetic rats, the up-regulated PAI-1 expression appears to take part in the process of the renal fibrosis.
