摘 要:目的探讨生远志及其皂苷与蜜远志对家兔离体十二指肠平滑肌的作用机制。方法采用离体肠肌实验,用生物机能分析系统记录生远志及其皂苷与蜜远志对正常肠肌及工具药乙酰胆碱、组胺、氯化钡所致离体肠肌收缩的影响,采集肠肌的振幅与频率,计算出各抑制率。结果生远志和远志皂苷均能显著抑制正常肠肌运动,其抑制率显著高于蜜远志(P<0.01);且生远志与总皂苷还能显著抑制乙酰胆碱(Ach)、组织胺、氯化钡所致的肠肌收缩,并与蜜远志相比有极显著差异(P<0.01);蜜远志对正常肠肌和氯化钡所致的离体肠肌兴奋性收缩也有抑制作用(P<0.05)。结论生远志及其所含的总皂苷对家兔离体肠平滑肌运动具有抑制作用;其抑制机制,可能系其与乙酰胆碱、组胺竞争,阻断M,H1受体以及直接抑制肠肌兴奋有关,且其抑制作用是不可逆的。蜜远志对正常肠肌运动有直接抑制作用,但显著小于生远志及其皂苷。Objective To approach the Crude Radix Polygalae and its saponin and Honey - stir - baked Radix Polygalae mechanism of in vitro intestine movement of domestic rabbit. Methods Used the in vitro intestine movement experiment, with the biologic function analyzer to record the influenses on ex- intestines and the intestines which contract were induced by Ach, Histamine and Barium Chloride ; Got the ex - intestines amplitude and the frequency, and calculated the inhibition ratio. Results Compared with the control, the Crude Radix Polygalae and its saponin could all remarkably inhibit the exintestines' movements, the inhibition ratios were remarkably larger than that of Honey -stir- baked Radix Polygalae( P 〈 0.01 ) ; and the Crude Radix Polygalae and its saponin could all remarkably inhibit the movements of the intestines which contract were resulted with Ach, Histamine and Barium Chloride ; the inhibition ratios were remarkably larger than Honey - stir - baked Radix Polygalae( P 〈 0.01 ) ; Honey - stir -baked Radix Polygalae also inhibited the excitability contract with the exintestines and the intestines which contract were induced by the Barium Chloride (P 〈 0.05 ). Conclusion ①Crude Radix Polygalae and its saponin can inhibit the ex vitro intestine's movement of the domestic rabbit ;the mechanism is possibly related with two factors, the one is Crude Radix Polygalae and its saponin compete with Ach and Histamine and block M, H1 receptor; the next is direct inhibition of ex vitro intestine excitability, and the inhibitory action is inreversible. ②Honey - stir - baked Radix Polygalae can directly inhibit the ex - intestine movement but the inhibitory action is remarkably less than Crude Radix Polygalae and its saponin.
