摘 要:目的通过豚鼠哮喘模型研究泡桐花总黄酮抗哮喘气道变应性炎症的作用。方法以卵蛋白(OA)致敏豚鼠为动物模型,灌胃途径予以泡桐花总黄酮大、小剂量,观察各组豚鼠支气管肺泡灌洗液(BALF)中白细胞总数、嗜酸性粒细胞(EOS)数和嗜酸性粒细胞特异性趋化因子(Eotaxin)的改变。结果泡桐花总黄酮可显著延长致敏豚鼠的引喘潜伏期,不同程度地抑制BALF中的EOS和白细胞总数及气道上皮Eotaxin的过度表达。结论泡桐花总黄酮对哮喘具有一定的保护作用,其中泡桐花总黄酮可能通过调控Eotaxin在气道黏膜上皮的过度表达,进而抑制Eos向哮喘气道的募集,消除气道炎症,成为其发挥抗过敏性哮喘的重要环节。Objective This study was designed to investigate the effects of flavonoids in Fructus Paulowniae on IAR and LAR. Methods An guinea pig asthmatic model, induced by OVA was administered by intragastric gavage(ig) with different doses of flavonoids in Fructus Paulowniae. The latent period, the number of acidophils and the Eotaxin in bronchoalveolar lavage fluid (BALF) were observed. Results The flavonoids in Fructus Paulowniae could delay the latent period of airway inflammation significantly. The number of acidophils and total white blood cell in bronchoalveolar lavage fluid(BALF) was restrained by flavonoids in Fructus Paulowniae. In addition, the flavonoids in Fructus Paulowniae could inhibit the overexpression of Eotaxin in the trachea epithelium. Conclusion The results suggest that flavonoids in Fructus Paulowniae can antagonize airway inflammation by inhibiting the overexpression of the Eotaxin in trachea epithelium.
