摘  要:目的观察乌体林斯(简称U)对哮喘患儿外周血Th1/Th2功能平衡的影响。方法采集20例哮喘发作期患儿和15例健康儿童外周血单个核细胞(PBMC),分别经植物血凝素(PHA)、PHA+U1(草分枝杆菌混悬液浓度为1×10-2g/L)、PHA+U2(草分枝杆菌混悬液浓度为1×10-3g/L)诱导后,测定培养上清液中白介素-4(IL-4)和γ-干扰素(γ-IFN)含量。结果哮喘患儿PBMC经PHA诱导后产生的γ-IFN、IL-4量与健康儿童相比,差别有显著性意义(P<0.01)。PHA+U1诱导组患儿PBMC产生的γ-IFN、IL-4量及γ-IFN/IL-4比值均明显高于单纯PHA诱导组及PHA+U2诱导组(P<0.01);PHA+U2诱导组患儿PBMC产生的γ-IFN量及γ-IFN/IL-4比值均明显高于单纯PHA诱导组(P<0.01),而IL-4水平则降低(P<0.05)。结论乌体林斯(草分枝杆菌混悬物浓度为1×10-2g/L)可明显促进哮喘患儿PBMC产生γ-IFN和IL-4,尤以前者为著,在促进Th细胞成熟的同时上调Th1细胞功能,改善哮喘患儿Th1/Th2功能失衡状态。Objective To study the effects of Utilin's on functional states of Th1/Th2 in peripheral blood mononuclear cells (PBMC) of asthmatic children. Methods Totally 20 asthmatic children and 15 healthy children were enrolled, the PBMCs of the subjects were collected and induced by PHA, PHA + U1 ( 1× 10^-2g/L) and PHA + U2 ( 1× 10^-3 g/L) respectively for 48 hours in vitro, then the levels of γ- IFN and IL - 4 in the supematants were detected by ELISA respectively. Results The levels of γ - IFN and IL -4 produced by PBMC stimulated by PHA of asthmatic children were statistically lower than that of the healthy children ( P 〈0. 01 ). The levels of γ - IFN and IL -4, and the ratio of γ - IFN/IL -4 in PHA + U1 induced group of asthmatic children were higher than those in PHA and PHA + U2 groups ( P 〈0. 01 ) , and the level of γ - IFN and the ratio of γ - IFN/IL -4 in PHA + U2 group of asthmatic children were significantly higher than those in PHA group ( P 〈0. 01 ), but the level of IL -4 in PHA + U2 group was lower than that in PHA group ( P 〈0. 05). Conclusion Utilin's ( 1× 10^-2g/L) can significantly increase the levels of γ - IFN and IL - 4 in asthmatic children, especially γ - IFN, and improve the functional imbalance of Th1/Th2 of asthmatic children by upregulating the function of Th1 cells based on promoting mature of Th cells.
