摘 要:利用压电细胞传感芯片(PCSC)对HepG2肝癌细胞的粘附行为进行24 h实时检测,获得经典响应曲线,根据HepG2肝癌细胞行为特性将响应曲线分为游离期、粘附期、平台期3个行为时段,并分析和计算每个行为时段的基本特性和平均用时;结合SEM实验和双胰酶消化实验,正反验证PCSC的主要参数因子-频率与细胞粘附行为信息之间的良好对应关系,分析所获得的数据,从定量分析的角度找到频率变化△f与粘附细胞数量间的数量关系,建立了以肝癌细胞为基础的压电生物芯片传感技术平台.Piezoelectric Cell-based Sensor Chip (PCSC) is applied successfully in the real-time monitoring of human HepG2 cell behaviors in vitro. The PCSC based on HepG2 cells is constructed and the classic curve is gained by the new technique, which is separated 3 phases, respectively the free phase, attachment phase and steady phase according to the property of the Frequency shift. The time spans and characteristics of the phases are analyzed. Using SEM to observe the adhesion of HepG2 cells on the gold crystal surface, different photos are obtained. In order to verify the cells behaviors, double trypsin assay is operated in PCSC. They reflect sensitive correspondence relationship between PCSC and cells'adhesion behaviors. There is a line relationship between the Af values and cell concentration attached on the surface of PCSC by analyzing data. The PCSC is constructed based on above experiments.
