摘  要:目的 探讨脊髓背角细胞外信号调节激酶(ERK)的激活在大鼠神经病理性疼痛诱发和维持中的作用。方法 雄性SD大鼠,体重200~300g。本研究分多剂量给药和单剂量给药两部分,均进行行为学实验和脊髓背角磷酸化ERK(p-ERK)表达检测。实验一 48只鞘内置管大鼠随机分6组(n=8):假手术+生理盐水(NS)组(S组)、慢性压迫性损伤(CCI)+NS组(C组)、CCI+5%二甲亚砜(DMSO)组(D组)、CCI+错义寡核苷酸10μg组(M组)、CCI+U01265μg组(u组)、CCI+反义寡核苷酸组10μg组(A组)。鞘内给药后记录大鼠机械缩足阈值(MWT)和热缩足潜伏期(TWL)。实验二 48只鞘内置管大鼠随机分3组(n=16),白CCI术前1d至术后3d分别注射NS(G1组)、U01265μg(G2组)或ERK反义寡核苷酸10弘g(G3组),假手术大鼠4只为阴性对照组,检测脊髓p-ERK的表达。实验三CCI术后5d大鼠40只随机分5组(n=8):CCI+5%DMSO组(D’组)、CCI+U01260.2腭组(U0.2组)、CCI+.U01260.5μg 组(U0.5组)、CCI+U01261μg组(U1组)、CCI+U01265μg组(U5组),假手术大鼠8只注射5%DMSO(S组)为阴性对照组,记录MWT和TWL;实验四另选CCI术后5d大鼠20只为实验组,鞘内注射U01265μg,假手术5d大鼠(阴性对照组)与CCI术后5d大鼠(阳性对照组)各4只,鞘内注射5%DMSO0.5h后取材,检测脊髓p-ERK的表达。结果 实验一与C组比较,U组与A组MWT和TWL升高,作用持续至术后13d;实验二与G1组比较,G2组与G3组术后3、5d胞浆p-ERK2表达以及术后3、5、10d胞核p-ERK1与p-ERK2表达降低;实验三与D组比较,U0.5组给药后MWT和TWL升高,其作用分别持续至鞘内给药后6h和2h;与U0.2组比较,MWT:U0.5组鞘内给药后0.5、2、6h、U1组给药后0.5、2、6、12h、U5组鞘内给药后0.5、2、6、12、24h升高,TWL:U1组鞘内给药后12h、U5组鞘内给Objective To investigate the role of extracellular signal-regulated kinase (ERK) in the spinal dorsal horn in the induction and maintenance of neuropathic pain. Methods Male SD rats weighing 220-300 g were used in this experiment. The experiment was divided into two parts: Part Ⅰ multiple doses and Part Ⅱ single bolus dose. In all animals PE-10 catheter was placed intrathecally with the tip located at lumbo-sacral segment of the spinal cord for intrathecal (IT) drug administration. Chronic constrictive injury (CCI) was produced by placing 4 ligatures on the left sciatic nerve at 1mm intervals with 4-0 silk thread. In Part Ⅰ 48 animals were randomly divided into 6 groups ( n = 8 each) : group S sham operation + normal saline (NS) 10μl (S) ; group M CCI + NS 10μl (C) ; group Ⅲ CCI + U0126 5μg in 10 gl (U) ; group Ⅳ CCI + 5% DMSO 10μl (D) ; group Ⅴ CCI + antisense ODN 10 μg in 10μl (A) and group Ⅵ CCI + missense ODN 10 μg in 10 μl (M). The drugs were injected through IT catheter twice a day for 4 consecutive days from the day before until the 3rd day after CCI. In Part Ⅱ on the 5th day after CCI 40 rats were randondy divided into 5 groups: group Ⅰ CCI + 5% DMSO 10μl ; group Ⅱ - Ⅴ CCI + U0126 0.2 gg ( Ⅱ ), 0.5μg ( Ⅲ ) ; 1.0 μg (Ⅳ) and 5μg ( Ⅴ ) in 10 μl. Eight rats were selected as normal control group :sham operation + 5% DMSO 10μl. The drugs were injected intrathecally only once. Pain threshold to yon Frey hair stimulation and thermal stimuli were measured, mechanical withdrawal threshold (MWT) and thermal withdrawal latency (TWL) were calculated before and after drug administration and the changes in MWT and TWL were determined. The expression of p-ERK was assessed by Western blot on the 3rd, 5th, 10th and 15th day after CCI in group C (NS), U(U0126 5 μg) and A (antisense ODN 10μg) in Part Ⅰ andat0.5, 2, 6, 12 and 24h after IT injection in group Ⅴ (U0126 5μ
