摘  要:目的从乳腺癌血管生成相关因子VEGF及MVD计数的角度探讨乳移平抑制不同乳腺癌肺转移的机理及模型间的差异。方法对BALB/c(SPF级)裸鼠和津白Ⅱ号(TAⅡ)小鼠分别采用人乳腺癌肺高转移细胞株MDA-MB-435和小鼠乳腺癌细胞株MA-891原位接种造模,随机分为乳移平组、环磷酰胺组和生理盐水组。每组小鼠给予相应药物4周后脱颈处死,称量记录瘤重,计算瘤重抑制率,计数肺转移结节数;免疫组化法检测VEGF蛋白,计数MVD。结果乳移平具有明显抑制不同乳腺癌肺转移模型肿瘤生长和降低肺转移发生的作用,可以降低不同模型肿瘤组织VEGF蛋白的表达,同时降低MVD数量,其中以对MDA-MB-435模型的作用更为明显。结论乳移平对不同乳腺癌肺转移模型均表现出抑瘤和抗转移作用,但对各模型的敏感性不同;乳移平对肿瘤组织VEGF蛋白表达的下调作用和降低MVD计数、抑制血管新生的作用与其抑制肿瘤肺转移关系密切。Objective To study the anti-metastasis mechanism of "Ruyiping Decoction" on expression of VEGF and number of MVD which are related to angiogenesis of breast cancer. Methods The MDA-MB-435 human breast cancer cells and the MA-891 mice mammary cancer cell cultured in vitro were subcutaneously inoculated into BALB/c nude mice or TAlI mice to establish the transplanted tumor model. The mice were then randomized into three groups and treated with "Ruyiping Decoction" and et al for 4 weeks. Then sacrifice the mice and weigh the tumor, calculate the inhibitory effect of tumor weight, count the number of the metastatic nodues in lungs, detect the change of VEGF protein and the number of MVD by immunohistochemical assay. The effect on MDA-MB-435 model is more obvious than MA-891 model. Results "Ruyiping Decoction" could inhibit the growth of tumor and depress the pulmonary metastasis conspicuously, lower the expression of VEGF protein of both tumor and lung, and the number of MVD. Conclusion "Ruyiping Decoction" can inhibit the tumor growth and pulmonary metastasis of different breast cancer pulmonary metastasis animal models, but its sensitivity to them is different. This Decoction's effects on down-regulating VEGF and MVD have close relation with its function of suppressing pulmonary metastasis.
