摘  要:目的:研究葛根素(puerarin,PR)与牛血清白蛋白(bovine serum albumin,BSA)的相互结合反应。方法:以 BSA 为荧光剂,PR 为荧光猝灭剂,在激发波长225 nm、发射波长340 nm 下测定荧光强度;分别测相同浓度的 BSA 和 PR 的荧光发射光谱和紫外吸收光谱。结果:随着 PR 浓度的增加,BSA 的荧光强度有规律地降低且呈良好的线性关系。结论:PR 与 BSA 的相互作用为静态猝灭过程。结合常数 K_R=1.37×10~6L·mol^(-1),供体(BSA)与受体(PR)间距离 r=3.44 nm,能量转移效率 E=0.670。根据热力学参数推测了 PR 与 BSA 的作用力类型。Objective: To study the interaction between puerarin (PR) and bovine serum albumin (BSA) in aqueous solution. Methods:Fluorescence was measured at 340 nm( emission wavelength) and 225 nm(excitation) with BSA as a fluorescence reagent, and PR as fluorescence quenching reagent. Fluorescence of BSA and UV - Vis absorption spectra of PR was measured at the same concentration. Results:The Fluorescence intensity decreased with increasing of PR's concentration regularly and had favorable action of them is a single static quenching process. The binding linear relationship. Conclusion:the combination reconstant is K = 1.37 × 10^6 L · mol^- 1. The shortest binding distance between BSA donor and PR acceptor is r = 3.44 nm and energy transfer efficiency is E = 0. 670. According to the thermodynamic parameters, the main type of binding force between them is suggested.
