摘  要:目的从细胞水平探讨缺氧预适应和前胡丙素对大鼠心肌细胞缺氧复氧损伤后胞内钙离子浓度与细胞凋亡的影响,从而阐明其保护心肌作用的机制。方法体外培养原代乳鼠心肌细胞,被随机分成5组:缺氧复氧损伤(HR)组、缺氧预适应(HP)组、腺苷预处理(AP)组、前胡丙素预处理(PP)组和维拉帕咪预处理(VP)组。在建立缺氧预适应和缺氧-复氧模型和用不同药物进行干预后,用全自动生化分析仪检测漏出LDH值。分别予FLUO-3/AM和Annexin-V荧光染色剂负载细胞后,利用流式细胞仪检测细胞内钙荧光强度和细胞凋亡率,并计算凋亡指数。结果同HR组比较,缺氧复氧组、AP、PP和VP组的LDH值、胞内钙荧光强度值和细胞凋亡指数明显降低(P<0.05)。而且PP组与其它干预组之间无显著差异性(P>0.05)。结论缺氧预适应、腺苷和前胡丙素预处理对缺氧复氧损伤后的心肌细胞均有保护作用,该作用可能与减轻细胞内钙超载,减少细胞凋亡有关;前胡丙素具有较强的钙拮抗作用。Objective TO explore the effect of hypoxia preconditioning and praeruptorin C on the concentration of intracellular Ca^2 + and the apoptotic index of cadiomyocytes in rats suffering from hypoxi-reoxygenation injury at the cellular level, so as to elucidate the mechanism of protective effect. Methods The neonatal rat cadiomyocytes were randomly divided into 5 groups: hypoxiarereoxygenation injury(HR) ; hypoxia preconditioning(H P), different medicines including Adenosine (AP), Praeruptorin C(PP) and Verapamil(VP). The value of LDH of the tran-sudate was detected by automaticbiochemistry analyzer after the two models were established. The fluorescence intensity of intracelluLar Ca2 + and rate of apoptosis in rat cadiomyocytes were detected with FLUO -3/AM and Annexin-V FITC loading with flow cytometer, and then apoptotic indexes were calculated. Results Compared with the HR group, the value of LDH, the fluorescence intensity of intracellular Ca^2+ , and the index of apoptosis were significantly low in groups of HP, PP and VP(P 〈0.05), but there was no significant difference in the pararneteres between the group PP and the groups of HP, AP and PP. Conclusion Hypoxia preconditioning and pharmacal preconditioning including AP and PP can produce the protective effects on cadiomyocytes suffering from hypoxi-reoxygena-tion injury, which may be related with lessening the intracellular calcium overload and reducing the apoptotic index of cadiomyocytes. In addition, praeruptorin C also plays the role of blocking Ca^2+ influxing into plasma membrane intracell in preconditioning.
