摘  要:目的探讨补体膜攻击复合物(membrane attack complex,MAC)对肾小管上皮细胞HK-2细胞外基质成分Ⅳ型胶原表达的影响。方法以纯化的c56及新鲜正常人血清(the normal human serum,NHS)作为C7~C9来源,体外组装sMAC建立HK-2亚溶破模型,通过流式细胞仪检测MAC刺激后的Ⅳ型胶原表达情况,荧光显微镜检测MAC的沉积。结果MAC刺激后的HK-2与对照组比较Ⅳ型胶原和合成增多(P〈0.05)。结论亚溶破剂量的MAC能使HKC的Ⅳ型胶原表达上调,可能是引起肾小管间质纤维化细胞外基质增多的原因之一。Objective To explore the effect of the membrane attack complex (MAC) on the expression of collagen type Ⅳ in renal tubular epithelial cells (TEC). Methods Purified C56 and C7 ,C8 and C5 come from the normal human serum (NHS) was added to form MAC in vitro, sMAC deposition was identified by immunofluorescence microscope. The expression of collagen type Ⅳ was assayed by FCM. Results Incubation of TEC with MAC increased the expression of collagen type Ⅳ (P〈0.05) in comparison with unstimulated cell. Conclusion MAC can increased the level of collagen type Ⅳ in cultured TEC, it may be among causes of abnormal accumulation extracellular matrix in TEC with tubulointerstitial fibrosis .
