摘  要:RNAi是一种有效的基因封闭工具,siRNA诱导RNAi效应的发挥。siRNA将哺乳动物细胞内特异基因的mRNA特异性降解而使基因失活,引起转录后基因沉默。siRNA有效作用的发挥关键是依赖它本身的性能及能否较高效率地转染入靶细胞后与靶基因结合。在体外常通过脂质体、聚乙烯亚胺及电穿孔等介导siRNA的转染。siRNA导入体内的过程则要复杂的多,到目前为止仍缺少有效的方法,常用脂质体和聚合物等来介导转染,依赖靶组织和靶细胞的类型来选用转导途径。RNA interference (RNAi) represents a powerful method for specific gene silencing. It is mediated through small double-stranded RNA molecules (small interfering RNAs, siRNAs), siRNA can induce special mRNA degradation. The special gene is interfered by post-transcriptional gene silencing. The critical factors that determine the success of RNAi have two, one is siRNA itself, the other is the ability to transfect intact siRNAs into target cells. The approaches to transfect siRNA in vitro usually use liposomes, polyethylenimine (PEI), electroporation, and so on. In vivo the delivery procession is more complicated and until now there are less efficiency methods. Liposomes and polycations are often used. The selection of delivery way relies on the types of target tissue and target cell.
