CD40和CD40L在大鼠炎症性肠炎外周血和结肠组织单-核细胞中的表达

摘　　要：目的:研究CD40和CD40L在大鼠炎症性肠炎(IBD)外周血和结肠组织单-核细胞(MC)中的表达与免疫功能紊乱的关系。方法:利用流式细胞仪对20例炎症性肠炎大鼠和20例正常对照组外周血、结肠组织MC上CD3、CD4、CD8、CD19的表达进行检测;CD40L在CD4+T和CD8+T细胞上的表达及CD40在CD19+B细胞上的表达进行检测。结果:IBD大鼠外周血、结肠组织CD3+、CD3+CD4+、CD19+细胞较正常对照组显著增高,CD3+CD8+细胞较正常对照组显著降低;IBD大鼠外周血、结肠组织CD4+T细胞和CD8+T细胞上的CD40L、D19+B细胞上CD40的表达都较对照组显著增高,且结肠组织上的表达又比外周血高。结论:CD40-CD40L途径在IBD免疫功能紊乱中可能起了重要作用。Objective： To investigate the relationship between abnormal expression of costimulator molecule CD40-CD40L on lymphocyte subpopulation and the immune dysfunction in IBD rats. Methods ： Samples from 20 IBD rats and 20 healthy controls were analyzed by flow cytometry to determine the expressions of CD3, CD4, CD8, and CD19 on lymphocytes, CD40L on CD4^＋ T cells or CD8^＋ T cells, and CD40 on CD19^＋ B cells on peripheral blood lymphocytes and Colon tissue of Inflammatory bowel disease（IBD） rats. Results： As compared with controls, the CD3^＋ T cells, CD3^＋ CD 4^＋ T cells, and CD19^＋ B cells on peripheral blood lymphocytes and Colon tissue of IBD rats increased significantly, but the CD3^＋ CD8^＋ cells decreased significantly; The CD40L expressed on CD4^＋ T cells and CD8^＋ T cells, and the CD40 expressed on CD19^＋ B cells on peripheral blood lymphoeytes and Colon tissue of IBD rats, were all significantly higher than those in healthy controls; expression of CD40,CD40L and CD4^＋/CD8^＋ on colon tissue was higher than on peripheral blood. Conclusion： CD40-CD40L pathway may play an important role in immune dysfunction of IBD .
