摘 要:目的研究抑制子κBα基因(IκBα)表达及其对激素敏感型单纯性肾病(SRSNS)病毒基因反式激活途径核因子κB(NF-κB)活性的调控作用。方法采用荧光定量RT-PCR、Western blot和电泳迁移率改变实验分别测定SRSNS活动期、SRSNS缓解期患儿和正常儿童外周血单个核细胞(PBMCs)中IκBα mRNA、蛋白质表达水平和NF-κB活性。同时采用RT-PCR和ELISA分别检测PBMCs呼吸道病毒基因表达和血浆病毒抗体。结果SRSNS活动期PBMCs细胞核中NF-κB活性增加,与SRSNS缓解期和正常儿童比较,差异有统计学意义(P<0.05)。NF-κB活性与呼吸道病毒检出率呈正相关趋势(OR=27,Kappa系数=0.59,P<0.05)。SRSNS活动期PBMCs细胞浆中IκBα蛋白质低于SRSNS缓解期和正常儿童(P<0.05)。SRSNS活动期NF-κB活性与IκBα蛋白质水平呈负相关(r=-0.884,P<0.05)。SRSNS活动期IκBα mRNA水平高于正常儿童,但仍低于SRSNS缓解期(P<0.05)。结论NF-κB介导的病毒基因反式激活过程中存在IκBα异常表达,IκBα对于NF-κB活性具有调控作用。Objective To investigate the expression of inhibitory kappa B alpha (IκBα) and it's regulatory effect on nuclear factor kappa B (NF-κB) during viral gene transaetivation triggered by respiratory tract viruses in steroid responsive simple nephrotie syndrome (SRSNS). Methods Children with SRSNS (including at active and remissive stage) were examined for this research project, and healthy children were chosen as controls. TaqMan reverse transeription-PCR (RT-PCR), Western blot, electro-mobility shift assays were performed to study the protein expression of IκBα gene and the activity of NF-κB in peripheral blood mononuelear cells (PBMCs). Simultaneously, RT-PCR was used to detect the gene expression of respiratory tract viruses (including respiratory syneytial virus and influenza virus), and viral antibody was measured by enzyme-linked immunosorbent assay (ELISA). Results Comparing with that from SRSNS at remissive stage and healthy children, the activity of NF-κB from SRSNS at active stage was much higher statistically (P〈0. 05). The positive correlation between NF-κB activation and the gene expression of respiratory tract viruses was observed (OR=27, Kappa coefficient =0. 59, P〈0. 05). In addition, levels of IκBα protein from children with SRSNS at active stage were much lower than those from healthy children and those from children with SRSNS at remissive stage (P〈0.05). There was a linear negative correlation between the activity of NF-κB and the protein level of IκBα (r=-0. 884, P〈0. 05). On the other hand, levels of IκBα mRNA from SRSNS at active stage were much higher than those from healthy children, but much lower than those from children with SRSNS at remissive stage (P〈0. 05). Conclusion Abnormal expression of IκBα, which is attributed to regulate activity of NF-κB, occurs during viral gene transactivation triggered by respiratory tract viruses in SRSNS.
