摘  要:目的:观察人类巨细胞病毒(human cytomegalovirus,HCMV)作用于血管内皮细胞(vein endothelialcells,ECV)对E选择素时序性的表达,探讨HCMV感染致动脉粥样硬化(atherosclerosis,AS)发生和发展的可能机制。方法:用HCMV感染ECV,采用RT-PCR方法检测不同感染时段细胞E选择素mRNA及蛋白的表达。结果:HCMV感染ECV后,感染0 h时E选择素mRNA有基础水平的表达,4 h升高并达高峰,8 h时开始回落,12 h回落显著,24 h回落更明显,与0 h比较,差异均有统计学意义(P<0.01),48 h接近0 h表达水平(P>0.05)。结论:HCMV感染ECV后能够促进E选择素表达,并且呈时间依赖性。HCMV有可能通过上调E选择素的表达,介导ECV的炎症反应,促进AS的发生、发展。Objective: By investigating sequential changes of E-selectin in human umbilical vein endothelial cells (HUVEC) by human cytomegalovirus (HCMV) infection, to study the potential mechanism of atheroscleresis(AS) induced by cytomegalovirus infection. Methods: HUVEC was cultured and then infected by HCMV. Expression of E-selectin mRNA was detected by reverse transcriptase polymerase chain reaction (RT-PCR). Results: Four hours after cytomegalovirus infection, the expression of E-selectin increasel. As the time of infection went by, the expression of E-selectin gradually decreased. When the cytomegalovirus infection lasted 48 hours, the expression of E-selectin returned to the fundamental level. There were significant differences among the groups at 0, 4, 8, 12, 24 and 48 hours ( P 〈 0.01 ). Conclusions: The infection with HCMV can enhance the expression of E-selectin mRNA in HUVEC. So we can say that HCMV may induce the inflammatory reaction by up-regulating the E-selectin expression on endothelial cells, which facilitates the occurrence and development of AS.
