摘 要:目的初步探讨HAART治疗后CD4+T细胞的CD44、CD95分子的表达水平变化。方法采用流式细胞仪检测CD4+处女型T细胞(CD45RA+T)和记忆型(CD45RO+T)表面CD44、CD95的表达水平,并分析二者的相关性。结果HIV-1感染组CD4+处女型T细胞(CD45RA+T)和记忆型T细胞(CD45RO+T)表面CD44分子的表达分别为21.17±8.36和28.40±10.49,比正常对照组的37.65±2.92和51.03±4.12低,具有显著性差异;与CD44分子的表达相反,HIV-1感染组CD4+处女型T细胞(CD45RA+T)和记忆型T细胞(CD45RO+T)表面CD95分子的表达分别为2.54±1.49和19.19±6.96,比正常对照组的1.51±0.11和13.68±1.37高,具有显著性差异。按照美国CDC对于艾滋病的临床分期标准对HIV感染者进一步分期,然后进行比较后,总体上仍反映出上述特点。另外,CD44和CD95的异常表达在B期尤为明显。经过HAART治疗以后,上述特点和异常能在一定程度上得到改变,但不能恢复,与对照组之间仍然具有显著性差异。研究还发现,细胞表面CD95和CD44的表达具有显著的正相关性。结论CD44分子的水平并不能代表体内病毒的拷贝数。HAART治疗后,新生的细胞在向淋巴结归巢的过程中,细胞凋亡增加。Objective To explore the changes of CD44 and CD95 expression on CD4^+ T lymphocyte of HIV-1 (human immunodeficiency virus-1 ) infected persons after effective highly active antiretroviral therapy (HAART). Methods Expression of CD44 and CD95 on CD4 ^+ CD45 RA^+ T and CD4 ^+ CD45 RO^+ T subsets were determined by flow cytometer in both HIV infection group and control group. Expression level of CD44 and CD95 was compared between the two groups. And relationship of CD44 and CD95 was also analyzed. Results The expression levels of CD44 on CD4^+ CD45 RA ^+ T and CD4 ^+ CD45RO ^+ T in HIV infection group were 21.17 ± 8.36 and 28.40 ± 10.49 respecfivdy, both of which were statistically lower than the control group (37.65 ± 2.92 and 51.03 ± 4.12 respectively). However, the expression levels of CD95 on CD4 ^+ CD45 RA^+ T and CD4 ^+ CD45RO^+ T in HIV infection group (2.54 ± 1.49 and 19. 19 ± 6.96 respectively) were both statistically higher than the control group (1.51 ±0.11 and 13.68± 1.37 respectively). When dividing the patients into A, B, C stage according to the stage standard of AIDS by US CDC, the tendency mentioned above was still the same. And the abnormal expression of CD44 and CD95 was most obvious in the stage B. After effective HAART, abnormality of CD44 and CD95 expression could be changed to some extent, but not restore to the normal level. It was also found a positive correlation of expression level between CD44 and CD95 . Conclusions Expression level of CD44 cannot predict the relative expression amount of the virus. Homing of remLscent CD4 ^+ T lymphocytes accompany with the increased apoptosis of themselves.
