摘 要:目的:观察低氧预适应对小鼠海马组织HIF-1与EPO的低氧应答元件(HRE)结合活性的变化,探讨这种变化与低氧预适应形成的关系。方法:小鼠低氧0次(H0),1次(H1),4次(H4)后取海马组织,应用凝胶迁移改变试验(EMSA),染色体免疫共沉淀(ChIP)试验和荧光定量PCR(real-time PCR)技术,检测小鼠海马组织内HIF-1与EPO的低氧应答元件结合能力的变化。结果:EMSA体外结合实验及ChIP体内结合实验发现,H0、H1和H4组结合活力依次增强。结论:HIF-1与EPO的低氧应答元件结合增强可能参与预适应的形成。Aim:To observe change of binding activity of HIF-1 with erythropoietin (EPO) hypoxia response element (HRE) in the hippocampus of mice preconditioned to hypoxia and explore relationship between the changes and the preconditioning. Methods: The hippocampus was removed from mice exposed to hypoxia for 0 run(control group), 1 run (H1 group) and 4 runs(H4 group). Electrophoretic mobility shift assays(EMSA), chromatin irrmaunoprecipitation( ChIP) and real time PCR were used to detect the change of activity of HIF-1 on HRE of EPO. Results: Both in vitro and in vivo binding tests showed that the HIF-1 DNA-binding activities were increased in group H1 and markedly increased in group H4. Conclusion: The increase of HIF-1 and HRE of EPO binding activities is thought be involved in hypoxic preconditioning.
