化瘀通脉注射液对大鼠主动脉球囊损伤后内膜增殖的影响

摘　　要：目的：观察化瘀通脉注射液对大鼠主动脉球囊损伤后内膜增殖的影响。方法：72只体重300～35％的雄性Wistar大鼠随机分为3组：即化瘀通脉组、丹参组和生理盐水组。采用球囊剥脱大鼠主动脉内膜的方法复制内膜增殖动物模型。自造模前1天开始，各组动物经尾静脉分别注射化瘀通脉注射液、或丹参注射液、或生理盐水，1mL／次，1次／d。每组按造模后1、5、10、15、20、25天6个时点随机选取4只动物处死，留取标本。制作病理切片，观察各组动物内膜损伤后内膜面积、内膜／中膜面积比值的动态变化，比较化瘀通脉注射液、丹参注射液及生理盐水对内膜增殖的影响。结果：光镜下（40×）观察病理切片可见，在行球囊内膜剥脱术后，3组动物主动脉内膜出现不同程度的增殖，内膜增殖随时间延长而加重，在术后25天最为明显，化瘀通脉组内膜面积增长缓慢，与生理盐水组比较有显著性差异（P〈0．05），和丹参组比较虽无显著性差异（P〉0．05），但增长有减缓趋势；3组动物内膜／中膜面积比值均随时间延长而增大，在术后25天最为明显，化瘀通脉组比值增大缓慢，与生理盐水组和丹参组比较虽无显著性差异（P〉0.05），但增大有减缓趋势。结论：化瘀通脉注射液可抑制大鼠主动脉球囊损伤后的内膜增殖，作用有优于丹参注射液的趋势。Objective： To study the effect of Huayutongmai Injection on intimal proliferation of rat aorta after balloon injury. Methods： 72 Wistar rats were divided randomly into three groups： Huayutongmai group （as experimental group ）, Danshen group （as positive control group） and normal saline group （as model group）. Four rats in each group, which aortic intima were injured by balloon to establish restenosis animal model, were killed and drawn materials in the lst,the 5th,the 10th,the 15th,the 20th,the 25th days after operations. Pathological changes of intima area and the ratio of intima to media area at different time were observed and the effect of Huayutongmai Injection, Danshen Injection and normal saline on intimal proliferatin was compared among three groups. Results： Different intimal proliferation was showed in three groups： the increasing of intimal area in Huayutongmai group was lower than that in normal sodium group （ P 〈0.05） and danshen group （ P 〉 0.05, but showed an obvious tendency） ; and the ratio of intima to media area in huayutongmai group was lower than that in the other two groups （ P 〉0.05, but showed an obvious tendency）, especially in the 25th day after balloon injury. Conclusion ：Huayutongmai Injection has a significant effect on inhibiting intimal proliferation of rat arota after balloon injury.
