血脉宁对血管内皮细胞p-选择素表达影响

摘　　要：目的观察血脉宁颗粒对血管内皮细胞(VEC)p-选择素(p-selectin)表达的影响,以探讨血脉宁保护血管内皮细胞的机理。方法以高脂血清、高脂血清加含药血清培养内皮细胞;ELISA法检测培养上清液p-selectin,流式细胞术检测内皮细胞表面p-选择素表达,RT-PCR检测p-selectinmRNA表达,Western-Bloting检测p-selectin蛋白。结果造模组血管内皮细胞表面p-选择素表达明显增高;血脉宁组内皮细胞表面p-选择素表达明显低于造模组。结论高脂血清可损伤ECV,增高ECV表面p-选择素表达;血脉宁含药血清可以使损伤的ECV表面p-选择素表达下降,具有保护损伤ECV的作用,其机理与其降低ECV表面p-选择素表达有关。Objective： To observe the influence on p - selectin expression, of Xuemaining Particles（XP, 血脉宁颗粒） on vascular endothelial cells, so as to investigate the mechanism. Methods： The serum was used to cultivate vascular endothelial cells （VECs） with the high - lipid serum or the serum containing XP. The p - selectin n the culture media were measured by ELISA. Flow cytomenter was used to measure p - Selectin on the surface of VECs, and RT - PCR to detect the mRNA of p - selectin. Results： There were the significant increase of p - selectin on the surface of the VECs in the normal control group. Compared to the normal control group there were distinct decrease of p - selectin on the surface of VECs in the XP group. Conclusion： The high - lipid serum can hurt VECs, which make the expression of the p -selectin increase. The serum containing Xue MiaNing could decrease the injuryed VECs to express p - selectin. XP had the effects of protecting the injury of VECs It was mechanisms to decrease of the p - selectin on the surface of VECs.
