摘　　要：目的检测基质金属蛋白酶（MMP）-2在胃癌组织中的表达及探讨其与淋巴结微转移的关系。方法对30例胃癌共850枚淋巴结采用CK-20mRNA RT-PCR扩增检测微转移，采用免疫组织化学染色检测该30例胃癌组织中MMP-2的表达情况。结果70％的胃癌组织MMP-2表达阳性；MMP-2表达与浸润深度、淋巴结转移和分化程度密切相关（P〈0．05），但与性别、年龄、肿瘤位置、直径、Lauren分型及淋巴管侵犯无明显相关（P〉0．05）；14例77枚淋巴结检出微转移；85.7％检测出淋巴结微转移的癌组织MMP-2表达阳性，高于未检测出淋巴结微转移的癌组织的56.3％。结论MMP-2表达与胃癌侵袭、转移和分化程度密切相关。MMP-2表达阳性可能参与淋巴结微转移的发生。Objective To examine the expression of MMP-2 in gastric cancer tissues and to evaluate its relationship with lymph node micrometastases. Methods The 850 lymph nodes from 30 patients with gastric carcinoma who underwent gastrectomy with lymphadenetomy were studied by using the RTPCR assay in addition to HE staining. Cytokeratin-20 gene marker was used in this assay. The MMP-2 expression in cancer tissues was examined by immunochemical technique （ EliVisionTM plus）. Results The MMP-2 expression was positive in 21 （70%） specimens. The MMP-2 expression in cancer tissues was correlated significantly with depth of tumor infiltration,lymph node metastasis, and tumor differentiation （P 〈 0. 05）. In contrast, no significant correlation was found between MMP-2 expression and other variables such as sex, age, tumor localization, size of the tumor, Lauren classifications and the lymphatic invasion （ P 〉 0.05）. Expression of MMP-2 was positive in 12 （ 85.7% ） of the 14 patients with lymph node micrometastases, and in 9 （56.3 % ） of the 16 patients without lymph node micrometastases （P 〉 0.05 ）. Conclusion The MMP-2 expression is strongly associated with depth of tumor infiltration,lymph node metastasis and tumor differentiation in gastric carcinoma. The positive expression of MMP-2 may contribute to lymph node micrometastases.
