摘 要:目的建立测定α细辛脑巴布剂中α细辛脑在家兔体内血药浓度的HPLC方法并考察其在家兔体内的药动学参数。方法采用消炎痛为内标,血浆经乙醚提取,采用C18(4.6mm×250mm,5μm)色谱柱,以醋酸盐缓冲液(pH4.5)-乙腈-四氢呋喃(40∶60∶5)为流动相,流速为0.8mL.min-1,检测波长为313nm,所得数据用3P97软件中的统计程序处理,求出有关的药动学参数。结果血浆中α细辛脑在0.05~2mg.L-1内线性关系良好(r2=0.9954)。平均方法回收率均大于90%,日内、日间精密度均小于5%。结论本方法简便、准确灵敏、重现性好,可用于α细辛脑的血药浓度分析及药动学研究。OBJECTIVE To establish a HPLC method for the determination of α-asarone cataplasm in rabbit plasma and to investigate its pharmacokinetic parameters in rabbits. METHODS The drug and the internal standard (indometacin) were extracted from plasma with ether, then detected by UV detecter at 313 nm with C18 column and sodium acetate buffer( pH 4.5 ) -acetonitrile-tetrahydrofuran(40: 60:5 ) as the mobile phase at a flow rate of 0. 8 mL · min^-1. The data obtained were processed with 3P97 program. RESULTS The linear range was 0. 05 - 2 mg · L^-1 ( r^2 = 0. 995 4). The mean recoveries of the assay were over 90%. Both the intra-day and inter-day validations were less than 5%. CONCLUSION The method is simple,sensitive,accurate and reproducible. It can be used for the pharmacokinetic studies of α-asarone in rabbits.
