摘 要:目的:建立以固相萃取-HPLC法测定人血浆中双氯芬酸钠浓度的方法。方法:色谱柱为ODS-2,流动相为磷酸盐缓;中液(pH6.5)-甲醇(40:60),检测波长为280nm,流速为1.0mL·min^-1结果:双氯芬酸钠血浆浓度在16.2~2024.0ng·mL^-1。范围内与峰面积积分值线性关系良好(r=0.9989),方法回收率在90%~110%之间,萃取回收率大于90%;日内、日间相对标准差小于5%。结论:本方法操作简便,结果准确可靠,可用于双氯芬酸钠药动学研究。OBJECTIVE: To establish an SPE(solid phase extraction) - HPLC method for the determination of diclofenac sodium in human plasma. METHODS: The separation of diclofenac sodium was performed on ODS- 2 column with a detection wavelength of 280nm .The mobile phase was composed of phosphate buffer(pH6.5) - methanol(40 : 60)at a flow rate of 1.0mL · min^- 1.RESULTS: The linear range of diclofenac sodium was 16.2-2 024.0ng· mL^-1( r= 0.998 9) .The methodological recovery ranged between 90% to 110% and the extraction recovery was above 90%. Both intraday RSD and inter - day RSD were less than 5% .CONCLUSION: The method is simple, accurate and reliable, and suitable for the pharmacokinetic study of diclofenac sodium.
