高剂量LH-RH对妊娠早期人滋养层组织蛋白分泌的影响

摘  要:本研究分析了妊娠早期不同周龄人胎盘绒毛滋养层组织培液中蛋白含量的变化。可知6、7、8、10周组织培养液,用SDS-聚丙烯酰胺凝胶电泳法,测得12种主要蛋白质,此12种蛋白的相对百分含量不同,其中有三种是随周龄增加而增加;有一种是随周龄增加而减少;另有七种蛋白在6周后,它们的百分含量维持在稳定状态;仅有一种蛋白从6周至10周几乎一直保持在稳定水平。有可能这些蛋白质在妊娠过程中起着不同生理作用,所以在不同周龄中的合成量亦有变化。本研究同时证明高剂量LH-RH对胎盘绒毛滋养层组织中分子量大约为50,000和6,000左右的蛋白有抑制作用。由此,讨论了这两种蛋白的消失是否与LH-RH终止妊娠的作用有密切关系。In this study the variation of secretory proteins by human trophoblastic tissue at 6-10 weeks of gestation grown in serum-free medium were analysed. The results indicated that there are 12 main proteins in the culture medium from 6-10 weeks' trophobkstic tissues. Three of them increased with the age of the trophoblastic tissues, one of the proteins decreased as pregnancy advanced and the another 7 proteins remained in constant level from 6th to 10th week. Based on the results that the variation of these secretory proteins in trophoblastic tissue as pregnancy advanced, we suggested that these proteins may play different physiological role during the early pregnancy.The experiments also indicated that the secretory proteins with molecular weight about 50,000 and 6,000 were inhibited by- LH-RH at a high dose of 50μg/2 ml culture medium. The relationship between the disappearance of these 2 proteins and the blockage of pregnancy by LH-RH is discussed.
