摘  要:研究了乙二醇(EG)、二甲基亚砜(DMSO)、丙二醇(PROH)和甘油(GLY)这4种渗透性冷冻保护剂及其两两组合对玻璃化冷冻未成熟(GV期)猪卵母细胞的影响;比较了3种不同冻前预处理方法和2种不同的解冻方法。结果表明:EG组卵母细胞所取得的成熟率为12.90%,与DMSO组(8.62%)及EG和DMSO混合组(9.61%)差异不显著(P>0.05),但明显好于其他各组(P<0.05);6步预处理法和3步预处理法所取得的卵母细胞存活率和成熟率显著高于1步预处理法(P<0.05),成熟率分别为12.33%和11.11%;3步和4步解冻法取得的成熟率要显著好于1步解冻法,成熟率分别达12.70%、10.47%。HUANG Jing et al (College of Animal Science and Technology, Northwest A & F Uniwersity,yangling,Shaanxi 712100) The present study was designed to examine the effect of four cryoprotectants and their mixture on the vitrification of porcine immature OOC3rtes and ~ three methods of pretreatment and two different dilution methods, The results indicated the effect of adding ethylene glycol(EG) was not significantly different from that adding dimethyl sulfoxide(DMSO) and mixture of EG and DMSO, but was significantly better than other groups (P〈0,05). And the maturation rates were 12,9%, 8.62% and 9.61% separately. The maturation of the one step pretreating method (1.49%) was not so well as those of the three and six steps pretreating methods (11.1% and 12.33%), The maturation rates of the three and four steps dilution methods (12.70% and 10.47% ) were higher than those of one step dilution method.
