摘 要:目的探讨药物性肝损伤的发病机理及保肝解毒颗粒的作用机制。方法60只小鼠随机分为6组,分别以相应剂量药物灌胃5天,再以雷公藤多苷灌胃造模,检测小鼠血清丙氨酸氨基转移酶(ALT)、血清门冬氨酸氨基转移酶(AST)和白介素18(IL-18)水平及肝细胞凋亡情况。结果模型组IL-18明显升高,肝细胞凋亡严重;保肝解毒颗粒可有效降低ALT、AST和IL-18水平,并使肝细胞凋亡程度明显减轻。结论雷公藤多苷所致的急性肝损伤与IL-18和肝细胞凋亡密切相关;保肝解毒颗粒可通过降低-18水平、抑制肝细胞凋亡等起到防治肝脏损害的作用。Objective To explore the pathogenesis of drug-induced liver injury and the therapeutic mechanism of Baogan Jiedu Granules. Methods Totally 60 mice were randomly divided into 6 groups. They were treated with intragastrical perfusion of corresponding doses of drugs for 5 days,respectively and then were modeled by intragastrical perfusion of Tripterygium Wilfordii Polyglycoside. Serum ALT,AST and IL-18 levels and apoptosis of hepatic cells were detected. Results IL-18 level in model group increased obviously with severe apoptosis of hepatic cells. Baogan J iedu Granules could effectively decrease ALT,AST activities and IL-18 level with apoptosis of hepatic cells obviously improved. Conclusion Leigongteng Duodai-induced acute liver injury is closely correlated with IL-18 and apoptosis of hepatic cells. Baogan Jiedu Granules has therapeutic effect for liver injury through decreasing IL-18 level and inhibiting apioptosis of hepatic cells.
