摘  要:本文建立了测定血浆中咪达唑仑浓度的反相高效液相色谱法。样品用缓冲液碱化后加氯仿提取。水浴挥干,加甲醇溶解进样。以C18化学键合硅胶为固定相,甲醇—水—乙腈(47∶38∶15)为流动相,安定为内标,在220nm波长定量检测,在5~500ng/ml浓度范围内线性关系良好,最低检测限为3ng/ml,方法重现性好,平均回收率为101.76%。用本方法测定7例单剂量输注0.12mg/kg咪达唑仑青年男性志愿者的血药浓度。A reversed-phase high performance liquid chromatographic method for the determination of midazolam in plasma is described in this paper. Diazepam was used as the internal standard. Midazolam was extracted from alkalized plasma with chloroform and evaporated to dryness. HPLC analysis was carried out onμBondpak C18 column with methanol-water-acetonitrile as the mobile phase and UV detection at 220nm. The lower limit of detection of midazolam in plasma is 3ng/ml, calibration curves is linear over the concentration range of 5 ̄500ng/ml. The assay procedure has been applied to monitoring plasma levels in intravenous infusion in 7 volunteers. The results show that the method is suitable for clinical pharmacokinetic research.
