摘  要:目的探讨急、慢性染铅对大鼠海马总量细胞外信号调节激酶2(ERK2)含量的影响。方法取正常30 d大鼠海马脑片培养,分别用含20μmol/L醋酸铅和不含醋酸铅的人工脑脊液(ACSF)孵育,作为急性染铅试验标本。大鼠在体染铅,取新生大鼠海马作为慢性染铅标本。用Western印迹法测定标本总量ERK2含量。结果急性染铅实验:染铅组和对照组总量ERK2含量均无明显变化。慢性染铅实验:慢性染铅总量ERK2含量于出生20 d后显著下降。结论急性染铅对总量ERK2含量无明显影响。慢性染铅使ERK2基因表达水平下降,总量ERK2含量明显减少。Objective To study the effect of acute and chronic lead exposure on total extraeellular signal regulated kinase 2 (ERK2) of rat hippoeampus.Methods For acute lead exposure in vitro ,20 umol/L lead acetate was added into the media.The control group in acute experiments used water instead of lead acetate. For chronic lead exposure in vivo, pregnant female rats were given 0.2 % lead acetate in their drinking water before breeding and throughout gestation and lactation. After weaning at postnatal 21 d,the pups were divided into control and lead groups 0.2 % lead acetate in drinking water. At 20,40,60,80 d,The rats in lead group were given hippoeampuses were got as chronic samples. The content of total ERK2 and phosphorylated ERK2 was detected by western blots. Results In acute experiment, total ERK2 of lead and control groups had no obvious changes. In chronic lead exposure, the content of total ERK2 decreased remarkably after 20 d. Conclusion Acute lead exposure may have no effect on the content of total ERK2. Chronic lead exposure can decrease the level of ERK2 gene expression.
