人单纯疱疹病毒Ⅱ型gD抗原的原核表达及免疫效果测定(英文)

摘  要:目的制备预防人单纯疱疹病毒感染的疫苗。方法扩增人单纯疱疹病毒Ⅱ型的gD基因,克隆入载体,导入大肠埃希菌表达系统,表达、分离、纯化抗原蛋白,然后进行小鼠动物免疫实验,测定小鼠的抗体水平;最后攻毒实验,观察小鼠的病理变化和死亡率。结果免疫后,gD抗原和IFN-γ联合免疫组小鼠体内产生抗体最高,其次gD抗原免疫组。生理盐水组小鼠病理变化出现最早也最重,且死亡最多;gD抗原免疫组小鼠病理变化出现较晚,症状也轻,死亡也少;gD抗原和IFN-γ联合免疫组小鼠无死亡。结论制备小鼠抗人单纯疱疹病毒的疫苗是可行的。此疫苗是否能刺激人体产生抗单纯疱疹病毒的抗体,是否对人有免疫效果,还有待于进一步研究。Objective To research vaccine to herpes simplex virus (HSV). Methods HSV-2 gD gene was amplified, cloned to vector and transformed to E. coli expression system. The product was separated, purified and injected into mice, then the Ab was detected. At last, the mice was infected by HSV-2, the pathological changes of mucosa and the death was observed, Results After immunity, gD and IFN-γ combined immune group produced the most antibody, gD immune group was in the second place. On the other hand, normal saline group owned the earliest and serious pathological changes, and the most death, gD immune group took the second place, while there was no death in gD and IFN-γ combined immune group. Conclusions Mouse antihuman HSV vaccine production is feasible. But it remained to be researched further whether it can induce human body to produce such antibody and have immune efficiency on hunman.
