摘 要:通过对粳稻9522γ射线诱变,从M2中筛选出1株带有白色中脉的隐性单位点突变的突变体,定名为Oswm(wm=white midrib)。Oswm叶片的近轴面中脉呈现白色,其他性状无明显异常。透射电镜观察结果表明,Oswm叶片位于白色中脉部位的叶绿体不能正常发育。为克隆OsWM基因,用Oswm突变体和籼稻广陆矮4号杂交的F2分离群体作为基因定位群体,通过图位克隆方法,首先将OsWM位点初步定位在水稻4号染色体上的SSR分子标记RM5503和RM2578之间。为了精细定位OsWM位点,在RM5503和RM2578之间发展了12个有多态性的InDel分子标记。最终将OsWM基因座位定位在CH413和CH415之间,遗传距离均为0.3 cM,物理距离约为122 kb。这为克隆OsWM基因以及研究叶绿体发育奠定了基础。A plant with a mutation of Oswm (wm = white midrib) was selected from a M2 population through mutagenizing keng rice cultivar 9522 with ^60Co γ-ray. The midrib on the proximal surface of Oswm leaf blade was white and other characters had not obviously abnormal. Transmission electron microscopy anal- ysis indicated that the development of chloroplast on the white midrib was abnormal. To map the Oswm locus, a F2 population generated by crossing the Oswm mutant with the hsien rice cultivar Guanglu' ai 4 was used as a gene mapping population. By means of the map-based cloning, Oswm locus was preliminarily located between SSR molecular marker RM5503 and RM2578 on rice chromosome 4. To further locate the Oswm locus, 12 polymorphic InDel molecular markers were designed. Finally, the Oswm locus was mapped between InDel marker CH413 and CH415. The genetic distances of Oswm away from these two markers were both 0.3 cM, and physical distance was 122 kb. These results are useful for further cloning and functional analysis of the Oswm .
