高碘对大鼠胸腺上皮细胞影响的超微结构观察

摘  要:【目的】研究摄入高浓度的碘化钾后,Wistar大鼠胸腺上皮细胞超微结构的变化。【方法】取断乳后1个月的Wistar大鼠,分为5组:适碘组(NI)、5倍碘组(5HI)、10倍碘组(10HI)、50倍碘组(50HI)和100倍碘组(100HI)。饲养3个月后,测定血清甲状腺激素水平,电镜观察胸腺上皮细胞的超微结构。【结果】与NI相比,10HI和50HI的FT3显著降低(P<0.05);100HI甲状腺激素的各项指标均显著降低(P<0.01)。电镜下高碘大鼠胸腺上皮细胞发生了明显变化:张力丝明显增多;溶酶体样致密颗粒明显增多;胞质空泡化;胞浆内出现许多低电子密度的囊泡;胞膜降解。尤其在100HI组,胸腺上皮细胞开始出现损伤性变化。【结论】不同部位的胸腺上皮细胞对甲状腺激素的反应不同;长期摄入过量的碘可使甲状腺激素减少,从而损伤胸腺上皮细胞。[Objective] To study the ultrastructural changes of thymic epithelial ceils (TEC) in condition of excessive iodine. [Methods] Wistar rats, 1 month after weanling, were divided into five groups, normal iodine group (NI), fivefold high iodine group (5HI), ten-fold high iodine group (10HI), fifty-fold high iodine group (50HI) and one hundred- fold high iodine group (100HI). Three months later, thyroid hormone was measured, thymic epithelial ceils were observed by electron microscopy. [Results] The FT3 level of serum thyroid hormone in 10HI and 50HI groups were lower than that in NI ( P 〈0.05); the levels of TT4, FT4, TT3, FT3, rT3 in 100HI decreased greatly ( P 〈0.01). Significant changes as significantly increased tonof'daments, numerous dense lysosome-like granules, many low electron density vacuoles degeneration of cell membrane and vacuolization could be seen in thymic epithelial cells by electron microscopy. [Conclusions] TECs of different location react to thyroid hormone differently. Excessive intake of iodine can destroy thymic epithelial cells by decreasing thyroid hormones.
