摘  要:目的从东海厚壳贻贝中分离纯化得到贻贝多糖MP-Ⅰ,研究MP-Ⅰ对四氯化碳肝损伤小鼠的保护作用。方法经DEAE-Sepharose、Sepharose CL-6B柱层析纯化获得贻贝多糖MP-Ⅰ。MP-Ⅰ对小鼠连续进行腹腔注射7 d,以0.2%CCl4橄榄油溶液经腹腔注射,建立小鼠急性化学性肝损伤模型,测定给药后模型小鼠肝指数、血清谷丙转氨酶(ALT)、谷草转氨酶(AST),肝组织匀浆超氧化物歧化酶(SOD)活性以及丙二醛(MDA)水平,光镜下观察肝组织病理学变化。结果得到MP-Ⅰ纯品。MP-Ⅰ高(100mg.kg-1.d-1)、中(50mg.kg-1.d-1)剂量组均能显著降低CCl4所致小鼠急性肝损伤血清ALT、AST活性,提高肝组织匀浆SOD活性,降低MDA,明显改善肝组织损伤的程度,与模型组比较差异有统计学意义,但对肝指数无明显影响。结论贻贝多糖MP-Ⅰ对四氯化碳所致小鼠肝损伤有保护作用。Objective To separate and purify polysaccharide (MP-Ⅰ) from Mytilus coruscus, and study the protective effect on CCl4-induced acute liver injury in mice. Methods The polysaccharide of MP-Ⅰ was purified using DEAE- Sepharose and Sepharose CL-6B column chromatography. The mice were treated with MP-Ⅰ via intraperitoneal injection for 7 days before CCl4 treatment. The activities of serum alanine aminotransferase(ALT )and aspartate aminotransferase(AST), hepatic superoxide dismutase(SOD) and malondialdehyde(MDA ), liver index, as well as the pathological changes of the liver were determined in all groups. Resuits We got pure polysaccharide MP-Ⅰ.The high(100mg·kg^-1·d^-1)and middle(50mg·kg^-1·d^-1)doses could significantly lower serum ALT and AST, hepatic SOD and MDA, and improve the liver histology. But it had no influence on the liver index of mice. Conclusion The polysaccharide MP-Ⅰ from Mytilus coruscus can protect the liver against damage induced by CCl4 inmice.
