摘 要:采用贴片法进行大鼠气管平滑肌细胞的培养,将不同稀释度的肺炎支原体培养液接种于单层细胞中孵育。对次日收集的细胞上清液进行肺炎支原体的PCR反应检测,结果为阳性,而冲洗液反应阴性,表明支原体对细胞具有较强的粘附性;接种后7d内观察到细胞产生明显病变,部分细胞发生崩解、脱落。MTT法检测结果表明支原体使细胞活性明显降低,其致细胞死亡率随滴度增高而增加,提示肺炎支原体对大鼠气管平滑肌可产生很强的致病性,使呼吸道受到严重损伤。本研究为探讨肺炎支原体致病作用机制及新药筛选与评价提供一种新的方法。The trachea smooth muscle cells in rats were cultivated by patch-adherence and the monolayer cells were inoculated by different dilution of the Mycoplasma Pneumoniae (MP) culture. The supernatant was collected in the following day and subjected to PCR analysis, MP was detected in supernatant, but not in the cell-flushing fluid, which indicated that MP had a strong adherence to the cells; Distinct lesion with some cells collapsing and falling off was observed 7 days after inoculation. The MTT assay showed that MP infection reduced cell cytoactivity with cell death increasing at higher dosages of inoculation. The results showed that MP had a strong pathogenicity to the trachea smooth muscle cells, causing significant damages to the respiratory tract. This study provides a new approach for exploring the MP pathogenic mechanism, and could be useful for the screening and evaluating of new drugs.
