补肾活血中药对大鼠骨髓间充质干细胞体外增殖的影响

摘　　要：目的观察补肾活血中药含药血清对大鼠骨髓间充质干细胞(MSCs)体外增殖的影响。方法采用Percoll密度梯度离心法分离纯化大鼠骨髓间充质干细胞,体外培养,建立大鼠骨髓间充质干细胞培养体系;收集补肾活血中药的含药血清,用10%含药血清代替10%的血清,绘制生长曲线,观察补肾活血中药含药血清促进骨髓间充质干细胞体外增殖的作用。结果所培养的大鼠骨髓细胞在表型表达和细胞形态上具备MSC特征,补肾活血中药含药血清组增长速度比空白对照组明显加快,并与剂量呈正比。结论补肾活血中药含药血清能促进干细胞在体外增殖。Objective To study the influence of the kidney - tonifying and blood - activating Chinese herbal medicine （KBCHM） on the proliferation of the SD rat bone marrow mesenchymal stem cells （MSCs） in vitro. Methods MSCs were isolated from bone marrow by density gradient centrifugation, then cultured in vitro. The 10 % serum containing KBCHM were collected, and then was used to take the place of 10 % serum. After the culturing, cell growth curve was drafted to evaluate the effect of the serum containing KBCHM on the proliferation of the rat MSCs in vitro. Results The cytomorphology and phenotypic expression of the cultured rat marrow cells had MSCs characteristics. The speed of proliferation in the group of containing the Chinese herbal medicine was more faster than that in the control group in a dosage - dependent manner, which proved that the serum containing KBCHM can promote the proliferation of MSCs in vitro. Conclusion Some Chinese medicines have cytoprotection function and can promote the proliferation.
