摘 要:目的研究川芎嗪对HaCaT细胞中FGF10mRNA转录和FGF10蛋白翻译的影响。方法HaCaT细胞以不同浓度的川芎嗪处理48h后,用原位杂交和免疫荧光法检测FGF10mRNA转录和FGF10蛋白翻译的改变情况。结果与对照组比较,处理48h后,FGF10的mRNA水平和蛋白水平均减少,随着川芎嗪剂量的增加,FGF10mRNA转录和FGF10蛋白翻译均减少,两组间比较,有显著性差异(P<0.01)。结论川芎嗪可抑制HaCaT细胞中FGF10mRNA的转录及其蛋白的翻译,随着川芎嗪剂量的增加,HaCaT细胞中FGF10的转录和翻译均减少。Objective To further illuminate the effect of the transcription of FGF10 mRNA and the translation of FGF10 protein in cultured HaCaT cells with tetramethylpyrazine treated. Methods HaCaT cells were treated with different concentration of tetramethylpyrazine for 48 h, then the transcription of FGF10 mRNA and the translation of FGF10 protein in these cells were detected by immunofluorescence and in situ hybridization assay. Results Compared with the control groups,with the concentration of tetramethylpyrazine increased,the transcription of FGF10 mRNA and the translation of FGF10 protein levels were deceased. There was significant differences among different groups (P 〈 0.01 ). Conclusion Tetramethylpyrazine could inhibit the transcription of FGF10 mRNA and the translation of FGF10 protein in HaCaT cells, with the dose of tetramethylpyrazine increased, the stains of both FGF10 mRNA and FGF10 protein in HaCaT cells reduced.
