摘 要:目的观察苦参碱(matrine)对体外培养人翼状胬肉成纤维细胞(human pterygium fibroblasts,HPF)增殖的影响。方法用噻唑蓝比色法(MTT)观察不同浓度(0.1、0.2、0.4、0.8、1.6、3.2、6.4、12.8mg/ml)的苦参碱在不同作用时间(24、48小时)对HPF增殖的影响。结果0.4~12.8mg/ml浓度范围内,作用24-48小时,苦参碱呈剂量依赖性的抑制HPF增生(P〈0.05)。结论苦参碱对HPF的增殖有明显抑制作用,并与用药剂量和持续时间有关。OBJECTIVE To study the influence of Matrine on the proliferation of cultured human pterygium fibroblasts (HPF) .METHODS Colorimetry MTr was used to detect viability of Matrine at different concentrations (0.1, 0.2, 0.4, 0.8, 1.6, 3.2, 6.4 and 12.8mg/ml) at different time (24, 48 hours) for inhibition of proliferation of HPF. RESULTS Administration of 0.4 - 12.8 mg/ml Matrine for 24 - 48 h could significantly inhibit HPF proliferation in a dose dependent manner ( P 〈 0.05) . There was obvious inhibitory effects of Matrine on HPFs in the concentration of 0.4 mg/ml for 24 h (P 〈 0.05, the rate of inhibition is 22.93 % ), and was more evident in the concentration of 0.4 mg/ml for 48 h ( P 〈 0.01, the rate of inhibition is 30.59 % ) as compared with control group.The 50 % inhibitory concentration (IC50) of Matrine was 1.8115 mg/mL.CONCILUSIONS There was a remarkable inhibitory effect of Matrine on HPFs, in a time- dependent and dose- dependent manner.
