摘  要:目的采用荧光金逆行示踪方法来评价自体骨髓间充质干细胞组织工程化神经修复狗坐骨神经缺损,再生神经的物质运输功能。方法在自体骨髓间充质干细胞组织工程化神经桥接狗坐骨神经5cm缺损,术后6个月,将5%荧光金(Fluoro-Gold,FG)注射入桥接侧的远段坐骨神经干内,2周后取相应的脊髓节段和背根节进行冰冻切片,并在激光共聚焦显微镜下观察结果,并进行了计量分析。结果桥接侧相应脊髓灰质前角和背根神经节切片中观察到被FG逆行标记的神经元。结论再生神经修复了缺损,恢复了神经干的连续性,再生神经具有良好的物质运输功能。Objective Used Fluoro-Gold to evaluate the axonal transport function of the regenerate nerve after a tissueengineered nerve of auto bone mesenchymal stem cells bridged to repair longer dog sciatic nerve gap. Methods Six months after grafting sciatic nerve across a 5 cm defect used a tissue-engineered nerve of auto bone mesenchymal stem cells, 5% FG solution was injected into the distal sciatic nerve trunk of the bridged side. Two weeks later, corresponding spinal segment and DRG were sliced on a cryostat and viewed under the confocal microscopy. Result FG-labelled neurons were found in both the DRG and the anterior horn of grey matter in the spinal cord ipsilateral to the bridged side. Conclusion The regenerate nerve repaired the gap, nerve continuity obtained restoration, and had a satisfactory axonal transport function.
