摘 要:目的探讨哮喘患者转化生长因子β_1l(TGF-β_1)基因多态性与血清TGF-β_1水平的相关性。方法采用聚合酶链反应—限制性片段长度多态性分析结合琼脂糖凝胶电泳的方法检测哮喘患者TGF-β_1基因型,采用酶联免疫吸附法测定血清中TGF-β_1水平。结果哮喘患者TGF-β-_1 TT型血清TGF-β_1水平明显高于CT型和CC型(p<0.01),CT型血清中TGF-β_1水平与CC型比较无显著性差异(p>0:05)。结论哮喘患者TGF-β_1基因多态性影响血清TGF-β_1水平。Objective To investigate the relationship between transforming growth factor β1 (TGF-β1) promoter polymorphism and serum TGF-β1 levels in asthmatic patients.Methods TGF-β1 polymorphism was detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Serum TGF-β1 was examined by sandwich enzyme-linked immunosorbent assay (ELISA). Results Serum TGF-β1 levels were significantly different in three genotypes of TGF-β1 in asthmatic patients (P 〈0.01). The TGF-β1 level in genotype TT was higher than that in the other two genotypes (P 〈0.01), but there was no significant difference between genotypes CT and CC(P 〉0.05). Conclusion TGF-β1 polymorphism is associated with TGF-β1 level in asthmatic patients.
