摘 要:目的观察甘肃黄芪异黄酮化合物对血管内皮细胞凋亡的影响及其可能机制。方法体外培养人脐静脉内皮细胞(HUVECs),将细胞分为对照组(A组)、血管紧张素Ⅱ(AngⅡ)(1μmol/L)组(B组)、AngⅡ(1μmol/L)+毛蕊异黄酮(1μg/mL)组(C组)、AngⅡ(1μmol/L)+2′-OH-3′,4′-二甲氧基异黄烷7-O-β-D-吡喃葡萄糖(1μg/mL)组(D组);荧光显微镜和激光共聚焦显微镜技术观察细胞的形态;Annexin-V/PI双染色流式细胞术检测细胞凋亡率及凋亡相关基因Fas蛋白的表达。结果B组与A组比较,B组HUVECs凋亡率、Fas蛋白表达率和平均荧光强度增加(P<0.001),激光共聚焦显微镜可观察到典型的凋亡细胞;C组与B组比较,毛蕊异黄酮抑制了AngⅡ的促HUVECs凋亡作用(P<0.001),降低了Fas蛋白表达率(P<0.001);两种黄芪异黄酮单体毛蕊异黄酮和2′-OH-3′,4′-二甲氧基异黄烷7-O-β-D-吡喃葡萄糖的组间比较,HUVECs凋亡率(P=0.195)和Fas蛋白表达率(P=1.000)均无显著性差异。结论黄芪异黄酮化合物可以抑制AngⅡ所致的体外培养HUVECs的凋亡。Objective To investigate the protective effects of isoflavones from Astragalus membranaceus on apoptosis induced by angiotensin Ⅱ (Ang Ⅱ ) in human umbilical vein endothelial cells (HUVECs) and its possible mechanisms. Methods Cultured HUVECs were randomly divided into four groups: control, Ang Ⅱ (1μmol/L), Ang Ⅱ (1 μmol/L)+calycosin (1 μg/mL), Ang Ⅱ (1 μmol/L)+2'-OH-3', 4'-dimethoxyisoflavanone-3-O-β-D-glucopyranoside (1 μg/mL). Mor- phology of apoptosis cells was observed by fluorescence microcopy with Hoechst 33258/PI double staining and laser scanning confocal microscope (LSCM). The expression of apoptosis related gene Fas was detected by FCM. Results Compared with the control, treatment with 1/μmol/L of angiotensin Ⅱ induced HUVECs apoptosis, the apoptosis ratio increased gradually (P 〈0.001),and Fas expression was upregulated (P 〈0.001). LSCM could distinguish apoptosis cells from normal cells. The injury effects of angiotensin Ⅱwere inhibited by the treatment of isofiavones from Astragalus membranaceus (P 〈0.001). When compared two kinds of isoflavones, no significant difference was found (P =0.195, P =1.000). Conclusion Isoflavones from Astragalus membranaceus has protective effects on HUVECs by inhibition of angiotensinⅡ induced apoptosis, suggesting that they may play an important role in prevention and treatment of vascular diseases.
