摘  要:目的:研究曲格列酮对THP-1巨噬细胞源性泡沫细胞ABCA1表达及胆固醇流出的影响。方法:采用液体闪烁计数法测定曲格列酮处理后THP-1巨噬细胞源性泡沫细胞胆固醇流出。用RT-PCR和Western blotting的方法检测曲格列酮处理后THP-1巨噬细胞源性泡沫细胞ABCA1 mRNA水平和蛋白质水平的变化。结果:经曲格列酮处理后,THP-1巨噬细胞源性泡沫细胞的胆固醇流出具有时间依赖性的增加,从0h的1.82%上升到24h的7.61%。在mRNA水平和蛋白质水平ABCA1也均随着曲格列酮作用时间增加而表达上调。结论:曲格列酮能增加THP-1巨噬细胞源性泡沫细胞ABCA1表达和胆固醇流出。Objective: To study the effect of Troglitazone on ATP binding cassette transporter A1 (ABCA1) expression and cholesterol efflux in THP-1 macrophage-derived foam cellss, Methods: After exposure of the cultured THP-1 macrophage- derived foam cells in Troglitazone for different times, cholesterol efflux was determined by liquid scintillator, ABCA1 mRNA and its protein level were determined by RT-PCR and Western blotting respectively. Results: The results showed that Troglitazone markedly increased ABCAl-mediated cholesterol efflux from THP-1 macrophage-derived foam cells, from Oh 1.82% to 24h 7.61%. This was accompanied by an increase membrane content of ABCA1, either in the level of protein or of mRNA. Conclusion: Troglitazone increases cholesterol efflux and the level of ABCA1 protein and mRNA in THP-1 macrophage-derived foam cells
