健脾软肝方及拆方对CCl_4肝纤维化大鼠MMP-2、MMP-9活性及蛋白表达的影响

摘　　要：目的:观察福尔肝健脾软肝方及拆方对CCl4肝纤维化大鼠MMP-2、MMP-9活性及蛋白表达的影响。方法:SD大鼠随机分成正常组、健脾软肝方及拆方组;复制CCl4肝纤维化大鼠模型,同时予等效剂量健脾软肝方及拆方灌胃,并以鳖甲软肝片为对照,观察血清肝功能;检测肝组织羟脯氨酸(Hyp),明胶酶图法观察组织MMP-2、MMP-9活性表达。结果:与正常组大鼠比较,模型组血清ALT、AST,肝组织中的Hyp含量与基质金属蛋白酶MMP-2、MMP-9活性及其蛋白表达都有显著增加。各组经相应药物治疗后,肝组织Hyp含量均有不同程度改善或降低,其中健脾软肝方全方组在降低ALT活性,肝组织Hyp含量较其他用药组为好,健脾软肝方全方组及部分拆方均能显著降低MMP-2、MMP-9酶活性;健脾软肝全方作用为显著。结论:福尔肝健脾软肝全方及拆方有明显抑制基质金属蛋白酶MMP-2、MMP-9活性及降低其蛋白表达作用,尤以健脾软肝方全方组尤为显著。此可能是福尔肝健脾软肝方抗肝纤维化作用机制之一。Objective： To observe theeffect of activity and proteinic expression of pharmacodynamics of the prescription of strengthening spleen and softening liver and separations on anti - liver fibrosis of big rat. Method： To randomly separatae SD big rat into three groups- normal, strengthening spleen and softening liver, separations; and copying the model of CCl4 anti - liver fibrosis of big rat, and give same dosage to the prescription of strengthening spleen and softening liver and separations to fill stomach simultaneity, and the the FuFangBiejiaruanganPian control group is comparison, and once a day all together 6 weeks; obsev the liver function of serum; detect the Hyp of liver tissue, and ob- serve the activity expression of MMP - 2, MMP - 9 by gelatinase picture. Result： Compare with normal group, the ALT and AST in model serum and the content Hyp of liver tissue and the activity and proteinic expression of MMP- 2, MMP- 9 had remarkably increase. After respectively medication, the content of Hyp in liver tissue improved or decreased in diverse level, among that the entire prescription group is better than others in reduce avtivity of ALT and the content of Hyp in liver tissue. And the prescription of strengthening spleen and soft liver and partly the prescription separations can decreased remarkably MMP- 2, MMP- 9 activity of enzyme, especially the therapeutic of strengthening spleen and soft liver. Conclusions： The prescription of strengthening spleen and soft liver and partly the prescription separations can decreased remarkably MMP - 2, MMP - 9 activity of enzyme, andthe therapeutic of strengthening spleen and soft liver was best. It is possible thant the above was one of the main mechanism of action of the therapeutic of strengthening suleen and soft liver on liver fibrosis.
