摘 要:目的探讨人工加载对悬吊和去势大鼠胫骨力学性能及力负荷对骨力学性能的影响。方法选取9月龄雌性SD大鼠,分为对照组、悬吊组、悬吊加载组、假手术组、去势组、去势加载组,采用自行研制的动态活体加载装置每日对悬吊加载组和去势加载组大鼠加载15min,频率5Hz,外部载荷3N。4w后处死大鼠,分别取胫骨皮质骨和松质骨进行横断面测量及压缩试验,分析胫骨的力学性能。结果悬吊组大鼠松质骨最大载荷及弹性模量较对照组明显下降(P<0.05),悬吊加载组最大载荷较悬吊组明显增大(P<0.05);去势组及去势加载组大鼠松质骨最大载荷及弹性模量较假手术组明显下降(P<0.05);悬吊组大鼠密质骨最大载荷及骨密质骨面积较对照组有所减小(P<0.05),去势组及去势加载组大鼠密质骨最大载荷较假手术组有所下降(P<0.05),去势组密质骨面积较假手术组有所减小(P<0.05),各组密质骨弹性模量无显著性差异。结论人工加载能显著提高悬吊大鼠胫骨的力学性能。去势大鼠骨重建阈值已经升高,需更大的负荷才能维持去势前的骨量与力学性能。Objective To investigate the effects of artificial loading on the mechanical performance of tibia of the suspended and ovarieetomized rats and to explore the effect of force loading on bone mechanical performance. Methods Forty two female SD rats aged 9 months were divided randomly into control, suspended, suspend & loading, sham operation, ovariectomized, ovariectomized & loading group. The tats in suspend & loading and ovariectomized & loading group were loaded in self-made dynamic in vivo device by 3 N and 5 Hz, 15 minutes.per day. 4 weeks later, the rats were killed to detect the cross section of dense and cancellous bone in tibia and do compression test to analyze mechanical performances of tibia. Results The maximum load and elastic modulus of cancellous bone of suspended group significantly decreased of control group ( P 〈 0. 05 ), those of ovariectomized and ovariectomized & loading group significantly decreaaed than thoae of sham operation group (P 〈 0. 05 ), The maximum load in suspended & loading group significantly increased than that in suspended group (P 〈 0. 05 ), that in dense bone of ovariectomized and ovariectomized & loading group reduced than that in sham operation group (P 〈 0. 05 ). The maximum load and dense bone area of suspended group decreased than those in control group ( P 〈 0. 05 ) , dense bone area of ovarieetomized group decreased than that of sham operation group (P 〈 0. 05 ), and there was no significant difference in elastic modulus of dense bone among these groups (P 〉 0. 05 ), Conclusions The artificial loading could enhance the mechanical performance of tibia of suspended rats, but for ovariectomized rats with the increased threshold value of bony remodeling, more loading may be needed to maintain the bone mass and mechanical performance before ovariectomy.
