两种小鼠胚胎移植方法的比较

摘　　要：目的:通过将传统的撕破囊膜进行小鼠胚胎移植的方法改为囊膜穿刺,以期提高胚胎移植的成功率.方法:共移植ICR和C57BL/6供体小鼠106只,其中改进方法移植47只(ICR小鼠31只,C57BL/6小鼠16只),传统方法移植59只(ICR小鼠39只,C57BL/6小鼠20只).结果:囊膜穿刺方法将输卵管和腹壁的炎性黏连发生率由60%降低到了20%,减轻了对移植后胚胎存活的不利影响,将ICR小鼠和C57BL/6小鼠胚胎移植成功率分别提高到了61%和37%.结论:通过囊膜穿刺进行胚胎移植提高了胚胎移植成功率.AIM： To promote the success rate of embryo transfer in mice by improving the traditional embryo transfer method. METHODS： Embryos were transferred to oviducts of 106 recipients by the traditional method （ tearing the bursa open over the infundibulum of oviduct） or the improved method （puncturing a hole in bursa with embryo transfer pipette and blowing embrys into oviduct through the hole）. ICR （n =31） and C57BL/6 （n = 16） pseudopregnant mice were transferred embryos by the improved method and other ICR （ n = 39 ） and C57BL/6 （ n = 20 ） mice did by the traditional method. RESULTS： The improved method reduced the blooding of bursa and the followed inflammatory adherence between oviduct and body wall, so it could achieve a relatively higher success rate of embryo transfer. CONCLUSION： The improved embryo transfer method can raise the success rate of embryo transfer in mice.
