更年甘露饮加小剂量雌激素对成骨细胞IL-1表达的影响

摘　　要：目的:观察复方中药更年甘露饮加小剂量雌激素对成骨细胞白细胞介素-1(IL-1)表达的影响,探讨一种治疗绝经期骨质疏松的新方法。方法:分别用模型血清、中药血清、中药血清加1/8、1/4、1/2最佳量雌激素及模型血清加最佳量雌激素用于第3代成骨细胞,用ELISA及免疫组化法检测成骨细胞IL-1的表达。结果:更年甘露饮加入1/8、1/2、1/4最佳量雌激素时均可降低成骨细胞IL-1的表达,加入1/4与1/2最佳量雌激素时作用更明显,与最佳量雌激素组相近。结论:更年甘露饮加小剂量雌激素可明显降低成骨细胞IL-1的表达,可作为一种治疗绝经后骨质疏松的方法。Objective： To investigate the effects of Gengnianganluyin plus low - dose estrogen on the expression of interleukin - 1 （IL- 1 ） of rat osteoblasts and to approach a new treatment to prevent postmenopansal osteoporosis. Methods： Osteoblasts were treated re. speetively with model serum, Chinese crude drug, Chinese crude drug plus 1/8 optimal estrogen, Chinese crude drug plus 1/4 optimal estrogen, Chinese crude drug plus 1/2 optimal estrogen and model serum plus optimal estrogen. IL - 1 expression was investigated by ELISA and immunohistochemistry. Results： The expression of IL - 1 was all decreased when rat osteoblasts were treated with Chinese crude drug plus 1/ 8 optimal estrogen, Chinese crude drug plus 1/4 optimal estrogen, and Chinese crude drug plus 1/2 optimal estrogen. The two later doses were more effective, which got close to model serum plus optimal estrogen. Conclusion： Gengnianganluyin plus low- dose estrogen can decrease IL - 1 expre.ssion of osteoblasts, which can be a new treatment to prevent postmenopausal osteoporosis.
