自体肿瘤细胞疫苗在结肠癌术后的主动免疫治疗研究

摘　　要：目的研究应用新城鸡瘟病毒修饰的自体肿瘤疫苗（NDV—ATV）对结肠癌术后患者机体免疫功能的影响，并探讨其临床意义。方法49例结肠癌患者术后第3周接种NDV-ATV（治疗组），采用流式细胞仪和酶联免疫吸附法，对其接种前后的外周血T淋巴细胞亚群、白细胞介索（IL）-2、IL—12和肿瘤坏死因子（TNF）-α进行检测，并以40例术后未接种NDV—ATV的结肠癌患者作为对照组。结果治疗组术后第6周CD^4＋T淋巴细胞计数和CD^4＋／CD^8＋比值明显高于术后第2周和对照组术后第6周（P均〈0．05）；治疗组术后第6周IL-2、IL-12、TNF—α较术后第2周和对照组术后第6周明显增高（P均〈0．05）。结论NDV—ATV是一种特异性较高的、安全有效的免疫制剂，能明显改善结肠癌患者术后机体的免疫功能，提高机体抗肿瘤能力。Objective To evaluate the availability of immune function recovery in postoperative patients with colonic carcinoma by Newcastle disease virus - modified autogenous tumor vaccine （ NDV - ATV）. Methods Three weeks after operation,NDV - ATV was inoculated in 49 cases of patients with colonic carcinoma. The serum level of interleukin （IL） -2,IL- 12 and tumor necrosis factor （TNF） -α expressions were detected with ELISA （ treatment group）, and selected 40 cases colonic carcinoma without NDV - ATV as control group. Results Six weeks after operation, the percentages of CD^4＋ cells in treatment group increased more significantly than those in 2 weeks after operation and 6 weeks after operation in control group. The serum level of IL -2, IL - 12 and TNF - α in treatment group also increased more significantly than those in 2 weeks after operation and 6 weeks after operation in control group （P 〈 0.05）. Conclusion NDV- ATV is a safe, effective and high specially immune pharrnaceuties, but it remains to be observed in remote effect.
