摘 要:为探讨疟原虫感染早期抵抗和易感小鼠Th1反应特点,以约氏疟原虫腹腔感染DBA/2和BALB/c小鼠为材料,用ELISA试剂盒和Griess实验分别检测感染第3天小鼠脾细胞、悬浮细胞以及巨噬细胞培养上清液中IFN-γ和NO水平。结果表明以PRBC或LPS刺激后,DBA/2小鼠脾细胞上清液中IFN-γ和NO水平随培养时间的延长均有不同程度的提高,但BALB/c小鼠48h培养组的IFN-γ和NO水平仅比24h培养组略有升高。两种小鼠单纯悬浮细胞或巨噬细胞上清液中IFN-γ或NO水平均明显低于其脾细胞上清液中的水平。这表明,约氏疟原虫感染早期,抵抗宿主对特异性或非特异性因子刺激的应答能力明显强于易感宿主,在相同因子刺激下前者建立的Th1反应可进一步被强化;巨噬细胞分泌NO明显依赖于IFN-γ的诱导效应。To clarify the Thl response characteristics of resistant and susceptible mice during the early phase of malaria parasite infection, the levels of IFN-γand NO in culture supernatants of splenocytes, suspending lymphocytes and macrophages from DBA/2 and BALB/c mice on the third day post-infection were measured. It was found that levels of IFN-γ and NO in splenocyte supernatants stimulated by PRBC or LPS from DBA/2 mice were remarkably higher than those of 1640 culture group, and they increased gradually with culture time. However, the levels of IFN-γ and NO in BALB/c mice splenocyte supernatants were unchanged after stimulation with PRBC or LPS. Both IFN-γ levels in suspending lymphocytes and NO levels in macrophages were significantly lower than that of corresponding splenocytes in two kinds of mice. The results suggest that immune response of resistant host to specific and nonspecific factors is obviously stronger than that of susceptible host during the early phase of Plasmodium yoelii infection, Thl response of resistant host can be further intensified by stimulus, and secretion of NO by macrophages depends on inductive effect of IFN-γ
