摘 要:目的建立从脑组织中分离人胚神经干细胞的方法和体外长期培养的稳定体系,为细胞治疗提供基础。方法采用机械酶学法从胎脑中获得神经干细胞,这些细胞在含成纤维细胞生长因子和上皮细胞生长因子的无血清培养基中生长,血清刺激诱导分化,程序冷冻法保存细胞。结果这些细胞可在体外持续增殖,并被诱导分化成神经元和神经胶质细胞,可以长期冷冻保存。结论人胚神经干细胞分离培养体系和冻存方法的成功建立为神经损伤、退行性疾病以及其他疾病的细胞治疗提供了潜在的细胞资源。[Objective] To establish a method for isolation, expansion and long term eryopreservation of multipotential stem cells from human fetal brains and provide a source for cell therapy. [Methods] Combination of enzyme digest and meehanieal dissoeiation were adopted to isolate stem eells from human embryonie eentral neural system (CNS), which can be maintained in serum free medium including epidemic growth factor (EGF) and b-fibroblast growth faetor (b-FGF), and indueed to differentiate by the serum and preserved by the eryopreservation programme. [Results] These eells were able to be expanded and indueed to differentiate to neurons or astrocytes in vitro and also ean survive after long term eryopreservafion. [Conclusions] The isolation and eryopreservafion system offered a potential source for eell replaeement in neurodegenerative diseases and brain injury or other diseases.
