摘　　要：目的 探讨human kallikrein 10（hK10）mRNA与蛋白在胃癌组织中的表达及其意义.方法 采用半定量RT-PCR、蛋白印迹法和免疫组化技术对41例手术切除的胃癌癌灶中心及其远端切缘中hK10的表达情况进行检测.结果 hK10 mRNA在胃癌癌灶中心和远端切缘组织中的相对表达量分别为0.21±0.20、0.02±0.03,两者相比差异有统计学意义（t=6.06,P＜0.01）.蛋白印迹法检测结果与其一致.癌灶中心的hK10阳性表达率为90%,远端切缘对照组为12%,两者相比差异有统计学意义（χ2=49.98,P＜0.01）.hK10表达与患者的性别、年龄、组织分级、临床分期、淋巴结转移无关（P＞0.05）.结论 hK10的表达异常可能与胃癌的发生有关,可能作为一种新的分子标记物在胃癌的诊断中起一定的作用.Objective To examine the expression of human kalikrein （hK） 10 gene in gastric cancer tissues. Methods The expression level of hK10 gene in the cancerous tissues proximal incisal margin in 41 samples was detected with RT-PCR. Western blot and immunohistochemisty methods. Results hK10 expression in cancer tissues was significantly higher than that in the proximal incisal margin as measured both at RNA level and protein level. No correlation was found between the expression of hK10 and clinical date such as patietns＇ gender, age, tumor location, histology, clinical staging and lymphatic metastasis （P 〉 0.05）. Conclusions The expression of hK10 may be associated with the carcinogenesis of gastric carcinoma.
