猪胸膜肺炎调查及地方适用型灭活疫苗的研制

摘　　要：对广东珠江三角洲地区31个猪场450份血清用IHA方法进行猪胸膜肺炎放线杆菌(APP)抗体检查,阳性猪场和阳性血清分别为96.8%和58.4%;分离鉴定了7株APP,血清分型为1、7、3和4型,且以1、7型毒力较强;用平板凝集法对263份APP抗体阳性血清进行抗体反向分型试验,结果1型、7型共占76.4%。确定了APP最佳液体培养条件,培养菌液CFU为2.4×109/mL;研制的APP 1型、7型二价灭活疫苗安全、稳定。兔和仔猪免疫抗体曲线显示,油乳剂疫苗明显优于铝胶疫苗,二次免疫优于一次免疫;APP双价油乳剂灭活疫苗一次免疫仔猪,能100%抵抗APP同源1、7型强毒菌株的攻击,保护期达110 d;田间试验明显提高猪群的生产成绩。Serological investigation was conducted for 450 serum samples collected from 31 swine farms in delta of the Pearl River by indirect haemagglutinin inhibition test （IHA）. The results showed that 263 serum samples derived from the 30 swine farms were positive, Seven isolates of Actinobacillus pleuroprteumoniae were successfully obtained, and classified into the serotypes 1, 7, 3 and 4, respectively, and the serotypes land 7 were more pathogenic. The 263 positive serum samples were further identified by plate agglutination reaction, and 76.4% of the samples were seropositive for serotypes 1 and 7. The concentration of cultured bacteria could reach 2. 4×10^9/mL after the most appropriate cultivation conditions were adopted. The bivalent inactivated vaccine were stabile and safe. The curves of corresponding antibody levels of groups of piglets and rabbits were detected post vaccination. The results showed that immune effects of the oil-adjuvant emulsified inactivated vaccine was better than that of the inactivated vaccine mixed with aluminium hydroxide gel, and the specific antibody levels of two inoculations were better than that of one inoculation, The protection rates of piglets inoculated with bivalent oil-adjuvant emulsified inactivated vaccine were 100% after challenge with virulent isolates of ActinobaciUus pleuropneumoniae serotype 1 and 7, and the duration of protective immunity was at least 110 days. Application of the inactivated vaccine in field-test demonstrated that more productivity could be obtained concerning vaccinated pigs in comparison with the control pigs.
